FlowCore Sorting/FACS Requirements

Sorting

If you are planning on sorting sterile cells, you need to have prepared your cells completely at your facility. That means having them in the correct tubes (Falcon 2063’s), filtered and at the correct concentration (Approx. 60-70 x 10^6 cells/ml.).  Cells can be transferred and filtered at the facility, but we make no guarantees of sterility, as we do not have access to a sterile room.

Please remember to bring with you: collection tubes, collection media, ice bucket (or something to hold the collection tubes in) and a PC formatted zip disk.

Controls required for your sort should include a negative, or unstained control, and single-stained positive control for each fluorochrome you are using.

If you are sorting into anything other than a tube, (96 well plate for example) please bring the plates preloaded with media, and an extra empty plate for instrument calibration.

If you are planning on sticking around during the sort, you may want to bring a book, or something to work on. 

Flow Cytometry Acquisition/Analysis Requirements

If you are doing FACS analysis, here are some things you may need to bring with you.

MAC formatted 250MB Zip disk. Data is backed up and archived, but we are not responsible for inadvertently lost data

Pipet and tips.  

**If you are doing PI staining for cell cycle, please make sure you have filtered your cells. Cells stained with PI tend to clump and will clog the FACScan. 

FACS Facility Rules

 We don’t like having a lot of unnecessary rules to complicate things, so please follow these simple rules:

1.  CLEAN UP AFTER YOURSELF. Anything left in the facility will become the property of the facility, or thrown away.

2.  CLEAN UP AFTER YOURSELF. To keep the instrument running consistently, please flush the system with bleach and water after your run.

3.  CLEAN UP AFTER YOURSELF. Please either take your trash with you or throw it away. There are trashcans in the lab, and they aren’t located on the benches.

4.  CLEAN UP AFTER YOURSELF. Actually that should read Respect Others Appointments, but I had a pattern going, and I didn’t want to break it. If someone is signed up after you, they have the right to ask you to finish up and stop. Remember, by delaying them, they may delay the next person, etc. etc.
