Submandibular Boood Sampling in Mice

Taking blood samples from mice using submandibular sticks

Equipment: Goldenrod Animal lancet 5 mm point size

Scruff the mouse, drawing the skin firmly at the nape of the neck very close to the head.

Take care that the mouse's head is in straight alignment with its body. The head cannot be angled down or to the side. Slightly compress the blood vessels of the neck on the contralateral side (opposite to the puncture site).

Locate the small, circular hairless area ventrally on the lower jaw and medial to the angle of the mandible. This hairless area may not be present in some strains of mice.

Puncture the skin with a 5 mm lancet slightly dorsally (1-2 mm) to this spot to a shallow depth of approximately 2-3 mm.

Blood will drip from this puncture site and can be collected in a centrifuge tube.

Blood will ooze at a rate of 1 drop per second. For a collection of 5 -10 drops (100-200 µl), the procedure takes nearly 15-20 seconds.

Once the collection is completed, release the mouse, and it will instantly stop the bleeding.
