My postdoctoral training took place in the laboratory of Dr. Daniel Tenen at Beth Israel Deaconess Medical Center, where I was engaged in a number of projects related to normal myeloid cell development and molecular causes of leukemia.  In one project, aimed to investigate regulation of the human CD34 gene, I found that the crucial cis element(s) are located in a remote position from the gene and efforts are currently under way to map them.  In a second project, I turned my attention to the role of CCAAT/Enhancer Binding Protein (C/EBP) family members in myeloid cell differentiation.  These studies identified C/EBPalpha transcription factor as a molecular switch of granulocytic differentiation.  Since C/EBPalpha function and/or expression was found to be perturbed in many cases of AML patients, I  designed a screen of a chemical library to find the compounds that upregulate C/EBPalpha expression, or increase its transcriptional activity, and may potentially give rise to new therapeutics for treatment of AML.  Finally, I am interested in modulation of C/EBPalpha function by phosphorylation, as a response to activation of various signaling pathways during leukemogenesis, and identifying protein partners of C/EBPalpha modulating its activity in phosphorylation-dependent manner.
