Major Research Theme
My laboratory is focused on several themes pertaining to platelet biology. First, we are interested in understanding the molecular basis of platelet granule secretion. To explore this problem, we have developed a permeabilized platelet model that we have used to identify molecules necessary for platelet granule secretion. Second, we have used a chemical genetic approach in order to identify signal cascades that are critical for thrombus formation in live animals. We have screened for and identified small molecules that inhibit platelet function and are presently assessing the effect of these compounds in an intravital mouse model of laser-induced thrombosis. These studies have lead us to evaluate the role of protein palmitoylation in platelet activation. Third, we are investigating the physiology of platelet microparticles. Specifically, we are determining whether platelet microparticles are derived directly from megakaryocytes and evaluating the mechanism of clearance of platelet microparticles. Finally, we are interested in using labeled platelets to visualize atherothrombotic disease in clinically relevant models. |
