Major Research Theme
1. TNF-related Apoptosis Inducing Ligand (TRAIL) has been shown to induce apoptosis in a variety of tumorigenic and transformed cell lines but not in normal cells, hence making TRAIL an ideal cancer therapeutic agent with minimal cytotoxicity. FLICE Inhibitory Protein (c-FLIP) is an important regulator of TRAIL-induced apoptosis.  We have demonstrated that persistent expression of c-FLIP(L) is inversely correlated with the ability of TRAIL to induce apoptosis in prostate cancer cells.  In a series of correlative and functional studies we have shown that persistent expression of c-FLIP(L) is necessary and sufficient to regulate sensitivity to TRAIL mediated apoptosis in prostate cancer cells.  Deciphering the molecular mechanisms of resistance to TRAIL can improve the efficacy of pro-apoptotic agents in treatment of malignancies.

2. A second project in the lab focuses on stromal-epithelial interactions in prostate cancer tumor progression. We have shown that fibroblasts associated with prostatic carcinoma can promote progression from preneoplasia to neoplasia. We are focusing on identifying the signal transduction pathways important in stromal-epithelial interactions in melignant and benign prostatic disease.
